急性肝衰竭小鼠肝组织IL-25和IL-17受体mRNA水平变化

Changes of IL-25 and IL-17RB mRNA in mice with D-galactosamine and lipopolysaccharide-induced acute liver failure

摘要: 目的 探讨白介素-25(IL-25)在急性肝衰竭发病中的作用。方法 采用D-氨基半乳糖和脂多糖混合腹腔注射法制备小鼠急性肝衰竭模型;采用实时荧光定量PCR法检测小鼠肝组织IL-25及其受体白介素17受体B(IL-17RB)基因信使核糖核酸(mRNA)水平;采用酶联免疫吸附试验法检测小鼠肝组织匀浆上清IL-25水平。计量资料采用t检验进行统计学分析。结果 肝衰竭小鼠肝组织IL-25和IL-17RB mRNA相对水平分别为(18.05±2.46)和(1.89±0.42),均显著高于对照组(P<0.05);肝组织匀浆上清IL-25蛋白表达水平为(26.17±14.00)pg/mg,也显著高于对照组【(12.50±4.68)pg/mg,P<0.05】。结论IL-25可能参与了急性肝衰竭的发生和发展过程。

Abstract: Objective To investigate the role of interleukin-25(IL-25) in acute liver failure in vivo. Methods An D-galactosamine and lipopolysaccharide-induced acute liver failure in mice was established. The relative levels of IL-25 and interleukin-17 receptor B(IL-17RB) messenger RNA in livertissues of mice were detected by real-time PCR and the IL-25 levels in homogenates of mice liver tissues was detected by enzyme-linked immunosorbent assay. Results The relative levels of IL-25 and IL-17RB mRNA in mice with acute liver failure [(18.05±2.46) and(1.89±0.4)] were significantly higher than in the controls (P<0.05);the levels of IL-25 in liver tissue supernatants of mice with acute liver failure were (26.17±14.00) pg/ml,significantly higher than that in the controls[(12.50±4.68) pg/ml,P<0.05]. Conclusion IL-25 may play an important role in the pathogenesis of acute liver failure.
