壳脂胶囊对NASH小鼠肝组织血红素氧合酶-1表达的影响

Effects of Kezhi capsule on hepatic expression of heme oxygeanse-1 in mice with non-alcoholic steatohepatitis

摘要: 目的 观察壳脂胶囊对非酒精性脂肪性肝炎小鼠肝组织血红素氧合酶-1(HO-1)表达的影响。方法 将C57 BL/6J小鼠20只随机分为对照组(采用蛋氨酸-胆碱充足饮食喂养)、模型组[给予高脂、蛋氨酸-胆碱缺乏(MCD)饮食喂养]、壳脂胶囊治疗组和自然恢复组。8周末检测血清转氨酶、瘦素水平,并观察肝组织脂肪变、炎症活动及纤维化程度;定量检测肝组织HO-1 mRNA的表达。结果 治疗组动物肝脂肪变及炎症程度较模型组明显减轻,血清瘦素水平和肝组织HO-1表达较模型组明显降低(P<0.05)。结论 在高脂、胆碱-蛋氨酸缺乏饮食诱导的NASH小鼠,壳脂胶囊可降低肝组织HO-1表达,阻止NASH的发生发展。

Abstract: Objective To investigate the potential role of Kezhi capsule on the hepatic expression of heme oxygeanse-1(HO-1)in mice with non-alcoholic steatohepatitis(NASH). Methods Twenty male C57BL/6J mice were randomly divided into four groups:mice fed with methionine-choline adequate diet were controls;The NASH model group were fed with MCD diet;The Kezhi capsule treatment group were fed with MCD diet for four weeks, then plus chitosan and the spontaneous recovery group were fed with methionine-choline adequate diet after four weeks MCD feeding. 8 weeks later,blood samples were collected for the detection of serum transaminases and leptin. The histological changes were observed under light microscope and HO-1 mRNA was detected by real-time quantitative PCR. Results Compared with those in MCD group,the hepatic pathological changes were ameliorated,and the hepatic expressions of HO-1 and serum leptin were decreased in Kezhi capsule group(P<0.05). Conclusions Kezhi capsule can down-regulate the expressions of HO-1 and leptin,so it can attenuate hepatic steatosis and necroinflammation in mice with NASH.
