HCV HVR1模拟肽诱导小鼠骨髓树突状细胞免疫应答机制的初步研究*

Preliminary study of immune response of dendritic cells in mice induced by a mimic peptide from HCV HVR1

摘要: 目的 探讨与丙型肝炎病毒相关的7肽诱导小鼠骨髓树突状细胞(DC)在体外分泌细胞因子的能力。方法 采用“Bulk培养法”从小鼠骨髓获得DC,经7肽负载;取经7肽免疫10天的小鼠脾细胞,分选获得CD4+T和CD8+T细胞;将脾细胞与DC共孵育24小时,收集上清,应用流式细胞术检测细胞因子。结果 培养的细胞表面CD11c 的表达率超过80%,具有典型的DC形态;实验组CD4+T细胞上清中TNF-α降低,IL-5、IL-2和INF-γ升高;实验组CD8+T细胞上清中IFN-γ、TNF-α、IL-10和IL-6较对照组有所升高,均具有统计学差异。结论 7肽能诱导以Th1及CD8+T细胞为主的细胞免疫反应。

Abstract: Objective To investigate the immune response and its mechanism of dendritic cells induced by a polypeptide. Methods Using the method of“Bulk”to harvest many DCs. After subcutaneously immunization for ten days,the mice splenocytes were harvested. CD4+T cells were separated from splenocytes by cell sorting technology. Then,CD4+T cells and DC cells were coincubated for a day. The supernatants were detected by cytometric bead array. Result The CD11c+ expression rate of DC cultured in vitro was more than 80%;Compared with control group,the level of TNF-α secreted by CD4+T cell was lower and the levels of IL-5,IL-2 and IFN-γ were higher in experiment group;the levels of IFN-γ,TNF-α,IL-10 and IL-6 secreted by CD8+T cell were increased in experiment group. Conclusion A large number of DCs from mouse bone marrow in vitro exhibites typical morphology of DC,which might be an experimental material for further study on function,markers and antitumor role of DCs in vitro. The 7 peptide could induce cell-mediated immune response through Th1 cells and CD8+T cells.
