杞蓟制剂对非酒精性脂肪性肝炎大鼠肝组织过氧化物酶体增殖物激活受体α mRNA的影响

The effect of QiJi preparation on mRNA expression of peroxisome proliferator activated receptor α in hepatic tissues of rats with nonalcoholic steatohepatitis

摘要: 目的 观察杞蓟制剂对非酒精性脂肪性肝炎(NASH)大鼠肝组织过氧化物酶体增殖物激活受体α(PPARα)mRNA表达的影响。方法 采用高脂饮食制备大鼠NASH模型,分别给予杞蓟制剂或复方蛋氨酸胆碱处理。采用原位杂交技术检测肝组织PPARα mRNA水平。结果 通过图像分析显示,与正常组(0.0457±0.0049)比,模型组大鼠肝组织PPARα mRNA表达的平均吸光度(0.0264±0.0036)明显减弱(P<0.01);与模型组比,杞蓟制剂和复方蛋氨酸胆碱防治大鼠肝组织PPARα mRNA表达的平均吸光度分别为0.0385±0.0042和0.0353±0.0037,水平明显增高(P<0.05),而杞蓟制剂与复方蛋氨酸胆碱处理组间无显著性差异(P>0.05)。结论 杞蓟制剂通过激活PPARα mRNA表达来调节脂质代谢,发挥防治NASH发生的作用。

Abstract: Objective To study the effect of Qi Ji preparation on mRNA expression of peroxisome proliferator activated receptor α(PPARα)in hepatic tissue of rats with nonalcoholic steatohepatitis. Methods The NASH model of rat was made by high fat diet. The rats were randomly divided into normal control,model,Qi Ji preparation and methionine/choline compound treated group. The mRNA expression of PPARα in hepatic tissues of rats was detected by useing in situ hybridization. Results By means of image analysis,the PPARα mRNA expression in model animals decreased(P<0.05)as compared with that in normal control group;QiJi preparation and methionine/choline compound increased the PPARα mRNA levels obviously. Conclution QiJi preparation could increase the PPARα mRNA expression in hepatic tissues.
